I have an 9'W X 24'L X 10'H room I want to use one 8" Vortex with a carbon filter. On the other end of the room near the floor I want to put an air intake with hepa filter. The room will be very tight. Do I need to have a fan on the intake or will the one 8" pull intake air.

If it's good and sealed you won't need a intake fan. If you want to balance the pressure some then u will want one. + I'm not sure how much the hepa would impede the flow, might require a fan to get the temps where you want them.

make the intake 2-3 times larger than the exhaust and you will be fine.
a passive intake will be easier because you want some negative air pressure. if a intake fan creates more airflow than the exhaust, the room will have positive pressure and some smell will escape out any cracks including the door when opened.
