As for the variation, I think this has a lot to do with the degradation effect. He states its effected by exposure to light and time.
Then he asks you to expose the plate to intense light by scanning it, so I think this is your loss.
I think its also best to use a digital camera no flash
I plan on using the plates to get an estimate of what's being carried over from one generation to the next
Yes its great to have an approximation on potency but I wouldn't say that's 100% accurate
Myself for THC potency I was looking at 5 samples all from the same plant not bud and compare for average
Keeping in mind to quickly get the picture of the dye exposure before it begins to fade.
So If I run 4 or 5 on a plate and they all have about the same size THC dot to measure then I have a good idea on what the THC% will range in
This is also a variable of grow nutrients & genetics I would hope that I am seeing more of a genetic variation since I don't follow a feeding regime
But only time will tell from the people who have chosen to test the plant and share the results
I also would like to keep a collection of the fingerprints
For example the one I called a Pink Pu$$y Hash Plant from the last grow showed similar traits to a Skunk Hashish Plant & Hashish Plant fingerprints

Again with strong THC THCV CBG & CBC variations.
I also learned from using these test kits that I prefer a strong CBG sedative effect.
I would say for the home tester that does not want to risk sending a sample out
This is a pretty effective way to track and monitor variants from genetic crosses
In addition to getting an approximation of THC content
Besides all of that isn't Science just FUN in general....
