I don't spray or mist but you are entirely right and could have potentially still got water on her .
The only thing I can think is when topping pot up she got a little dirty , had to blow a bit loose dirt off of her but don't see how that could cause it :S

Flip out the lights for maybe 15-20 mins and give her a nice foliage spray, maybe add a pinch of silica as well, make sure the foliage spray is at least 6.8+ to help with calcium uptake. The proplem here is it looks more like a mag problem than a cal problem. Both are very similar and mag requires more of a acidic pH and cal requires more of a base pH, that's why I say spray first then hit it with mag if you don't see it green up in a matter of hoursoh, and none of that is for the tiny yellowing only on that leaf.
that's just a genetic abnormality seen it probably 50 times at least.

100% certain it's not a deficiency due to cal or mag, too early for those to manifest.
container is too big for that plant as well, which is contributing to your first problem... too wet of a media.
when you water, just dribble some around the base of the plant rather than soaking the entire container.
after it's established then you can water the entire container, but only after the plant grows more, approx. double the growth you see now.
for now, scrape or simply agitate the surface of the media to allow more water evaporation.
a fan couldn't hurt either.
and make sure you have that plant OFF the floor, you want something under it so it doesn't sit in any drainage at all

also important to remember that when rain falls it mixes with co2, which makes it more acidic naturally, and that ph is fooling.
