Tissue Culture : Small Indoor Setup

Hey Everyone!

Curious if anyone has dabbled in the tissue culture sector. I know it's been around for decades but more so in cannabis in the recent years.

I am interested in setting up a clean room and tissue culture lab but am curious as to the benefits of going tissue culture route & also if anyone has set up a room for tissue culture maybe they can help me.

It's been on my to do list for awhile. Just haven't gotten around to it as I have so much other stuff going on.

I've been looking at some of the kits on this site but haven't pulled the trigger.

It's an adventure for sure !! I have been going to @Test-tube-garden they were recommended to me by a fellow homie. Have you started getting anything in your kit ?

I haven't purchased anything as of yet. I don't really have the space or time to get started with tissue cultures and when I do I want to do it right. So until I can devote a decent amount of time for more research it's on hold for now. But it is something I'm definitely going to pursue in the future.

How long can you hold on to a culture? Does a rooted culture count as a plant in Michigan?

So I too am curious how long the tissue cultures would last, I have seen how fungi can be suspended in tissue culture for years essentially due to the cold slowing down its cellular division. With a cannabis clone I read that as a plant it would need to be re-cultured every few weeks to keep it alive. Would putting a tissue cultured herbaceous like cannabis in cold storage not damage the plant as it does not have any dormant periods? I wanted to tissue culture for genetic preservation but it seems that its more of a way for plant propagation on a bigger scale for someone looking to really make a lot of clones faster than normal. On a small or even medium scale it seems like regular cloning is most effective for how easy it is to cut almost any sized cut, apply an auxin, place in wet media, bing-bang-boom 1-3 weeks you're transplanting 90-100% cuts into new homes. Tissue culture you gotta mix up the solution carefully, or buy just the right solution, sterilize EVERYTHING and BEYOND, and for what?...you shave a few days off rooting time?...I must be missing something...I think tissue culture is also the type of application that is for stuff like orchids that have serious issues with normal germination because their seeds don't have an endosperm to feed the embro, which cannabis has plenty of in healthy seeds. But if anyone knows or has experience with how they store tissue cultures for long periods without having to re-culture wvery few weeks...I'm all ears for real

I tried this years ago.

I found it next to impossible (monetarily speaking) to create an appropriately sterile environment necessary for soft tissue propagation. All it takes is ONE fungal spore making its way into the culture media to ruin hours of hard work. Getting the right auxin to cytokine ratio for the appropriate "rooting" response was also difficult. It requires a lot of time/trial and error.

I wouldn't suggest trying this at home kids...

Laminar flow hood, industry standard filtration %99.99 down to .3 microns, been doing sterile lab work with them for 10+ years. would love to media recipes from one who has experience with tissue culture of cannabis

This is on my radar as well. I want to try using my own clones but I don't have a separate grow room to keep mothers so I thought this might be a way to get around that. What conditions are needed to store the samples (light/dark/temperature)?

Mid 70's, I used fluorescent, 2 t1 per shelf four or five inches up.

12 12 seemed OK.

What will hold you up is rooting and hardening off.

Have 0% experience with tissue culture / micropropagation. How long are clones in agar stable for shelf life wise?

Dude your killing it.
I'm currently having trouble with sterilizing my explant material. But im a budget friendly guy and I have been getting success with making my media and not getting contamination. But I'm failing with the explants. They brown out on me and die or show contamination on the plant and moves to the media. Like I said I'm a budget guy and working my way into it.
